Protocol

  1. Preparation: Resuspend PBMCs in separation buffer.
  2. Labeling: Add the Isolation Cocktail and incubate for 15 minutes at 2-8°C.
  3. Washing: Wash cells with buffer to remove unbound antibodies.
  4. Bead Incubation: Add Anti-Biotin Nanobeads and incubate for 15 minutes at 2-8°C.
  5. Separation: Pass the suspension through the LS Column; collect the flow-through (enriched NK cells)

FAQ

Q1: What are the sample requirements? Can I use frozen PBMCs?
The kit is optimized for human PBMCs. Both fresh and cryopreserved PBMCs can be used. However, for frozen samples, ensure high viability (>90%) and remove clumps/debris with a strainer before labeling to prevent column clogging.
Q2: Since this is an Untouched selection, can I stain the cells immediately after isolation?
Yes. Because the NK cells in the flow-through are "untouched" (no antibodies or beads bound to them), all surface receptors are available. You can immediately proceed with flow cytometry staining or functional activation.
Q3: How does the Column-based separation work?
The column contains a matrix that amplifies the magnetic field. Magnetically labeled non-target cells are retained inside the column, while the unlabeled (untouched) NK cells pass through freely.
