Application
Protocol
- Preparation: Prepare a single-cell suspension from Human PBMCs.
- Labeling: Add CD3 Microbeads directly to the cell suspension. Incubate to allow binding to CD3+ T Cells.
- Separation: Apply the suspension to a separation column placed in a magnetic field.
- Washing: The labeled CD3+ cells are retained in the column, while unlabeled cells (B cells, NK cells, Monocytes) are washed away.
- Elution: Remove the column from the magnetic field and elute the highly purified CD3+ T Cells.
Validation Data
Efficient Isolation of Pan-T Cells Using SOLIDEX®-ISOEx Human CD3 Isolation Nanobeads (Column-Based)
Catalog No.GM-Tg-hg-T87075-iso-nanoIMB
To assess the purity of isolated T cells, CD3+ T cells were magnetically sorted from human peripheral blood mononuclear cells (PBMCs). Cells before and after separation were stained with APC-conjugated CD3 antibody and propidium iodide (PI), followed by flow cytometric analysis. The results demonstrated a significant increase in CD3+ T cell purity from 52.6% pre-isolated to 95.0% post-isolated, confirming successful T cell isolation.
Figure 7. Efficient Isolation of Pan-T Cells Using SOLIDEX®-ISOEx Human CD3 Isolation Nanobeads (Column-Based). The T cells were labeled with SOLIDEX®-ISOEx Nanobeads conjugated with anti-CD3 antibody. And then these cells were enriched by using cell isolation column. The T cells in pre-isolated human PBMCs. T cells with high purity and motility were enriched.
Background
Why Choose SOLIDEX®-ISOEx Human CD3 Isolation Nanobeads?
- Superior Recovery: Positive selection strategy ensures the capture of the maximum number of target CD3+ T cells, ideal for rare samples.
- High Specificity: The column-based washing step efficiently removes background cells, delivering exceptional purity.
- Biodegradable Nanobeads: The tiny magnetic beads are non-toxic and biodegradable, minimizing interference with cellular physiology.
- Fast Protocol: Simple incubation and separation steps allow for isolation in approximately 45 minutes.
